Sorry I don't get it, does it mean that the offer duration is at least a year or I should have been already working for a year i.e. I cannot claim the points before spending a year in my job? Also, what does this sentence mean; for at least one year after we issue your permanent resident visa?

Yes, you could go to any ASC within the USA without an appointment. You just need to take the letter of biometrics request with you.

Hi , I think all work permit applications from USA are processed at the Consulate of New York. Processing time currently is 3 months on average. You can also follow their page on facebook, they post weekly processing updates.
