Depends.

What info are you missing in your reference letter and is it included in the contract?

I'm missing the main duties . My contract contains a job description so a list of all the duties and responsibilites .
it is signed by HR but doesn't have a proper letterhead , it has the name of the company on the top .

Without duties matching to specific NOC, there is a good chance the letter wont work.

If you have the recent letter from HR - any chance that the letter references the contract? Also, any chance that you have your employer's advice to attach the contract in writing (e.g. an email)? You could write a letter of explanation and include any proof you have of her refusal to provide that.

But if you have the general letter, plus the contract, plus any supporting documents, and the contact information of the person writing the letter, that might be enough. Do you have a colleague who could also write a letter?
