I have applied for PR. I got overall 6 bands, but 5.5 in two modules (Writing and Speaking) . Should I reappear for the exam? Will this affect my visa application? Is it mandatory for spouse to appear for the exam?

Not sure how you applied for PR, your 5.5 in the two modules would be ineligible. You need CLB 7 in all modules to submit an Express Entry profile. Not mandatory for spouse.
