I had a previous marriage last for 12 years I got divorce then got married again ..There is a 16 years different between my wife and I but the relationship is genuine. . would be that problem for the immigration to decide to refuse the sponsor ship application?

not if you include proof of your divorce (like, the legal papers) and provide sufficient proof of the genuine-ness of your new relationship.

I think it will only affect your chances if you applied for PR for your former spouse and had legal trouble/fraud/problems in the immigration process. Then applying for PR for your new spouse might raise some red flags. Otherwise, no - a divorce in your past will not cause problems for your immigration, although you do need to provide all the legal documentary proof of the divorce as well as proof of any custody plans etc. for children from that marriage.
