I have completed both Bachelors and Masters Degree. Unfortunately I missed one of my marksheet of Masters Degree (dont have consolidated Marksheet) due to which I may not be able to do ECA for that degree in the near term. :( Can I create a Express Entry profile with my bachelors degree as of now? Later is there a possibility to add my Masters degree into the profile once I get a duplicate Marksheet and ECA has been completed. I completed my masters degree at Anna university. Whether anybody used the tool itranscripts of Anna university? Whether we need all Marksheets to apply for the transcripts or any alternatives for the same? Please give in your suggestions. Thanks for your help and support.

You need ECA evaluation to create an express entry profile. Is your bachelor's degree evaluated already? If you have an ECA for your bachelors degree then you can go ahead and create a profile for now but if there is no ECA, you won't be able to do it.

You need only consolidated mark sheet to apply for transcript through itranscripts. Individual semester mark sheets not required. I used itranscripts for my spouse transcript, my experience was good. They promptly respond your queries and once their document team approves your photocopies , you get the transcripts real quick. But the only thing they missed was that they didn't mention that Anna university doesn't provide mark sheet attestation for autonomous college (psg, gct, cit types) mark sheets. They just sent the degree attestation to Wes (I chose direct Wes sending) and Wes requested for transcripts further. It wasted a lot of time for me. But if you haven't studied in those colleges, I believe you are good to go with them. While choosing the options in itranscript, just choose degree + mark sheet one. You don't need Wes form and waste not extra 400 on that. You can choose your mark sheets directly sent to Wes through itranscipts or you can receive it and send too. I'd prefer latter because you can be sure of what goes to Wes and avoid scenarios like what happened to mine.

See if you are getting 440+ with bachelors alone. Otherwise don't bother much on creating your profile in a hurry. Also Wes has one charge to assess both bachelor and master degrees, so if you create separate application for both, you'd waste another $250+ unnecessarily.
