Hi

My criminal rehabilitation application is recently approved by Canada consulate in India. I have been given an approval letter as well by the consulate

Now, I am planning to apply for work permit through my employer. Am I still required to disclose the conviction from USA for which I am rehabilitated on work visa form IMM 1295E under background information section ? Does approval of criminal rehabilitation permit me to no longer disclose it on Canada visa application ?

Yes - you are still required to disclose them.

All the rehabilitation application does is allow you to enter Canada. It doesn't make these convictions go away.
