I am sponsoring my wife (in-Canada) for Permanent Residency and am familiar with the fees, EXCEPT if there is an additional fee for 5710E (the permit to allow her to work while the Permanent Residency process is occurring). I've read the Work Permit Guide, but can't figure out.

My wife is Brazilian. Here in Brazil there is a Permanent Visa for spouses. As far as I have read, there is no intermediate step like this before Permanent Residency in Canada. My wife would have the 180 days (six months) that she has on her Tourist Visa (and she might be working with the Work Permit), but staying in Canada beyond this 180 days during the 12 month Permanent Residency process would be prohibited (unless the Work Permit is extended, which might be an option? or have I missed something?

Either she is a visitor or a worker. If she is a visitor, she can extend her status. If she is issued an OWP, it should be valid for 2 years.

No prob. I should have mentioned that the OWP with the inland PR app should be valid for 2 years unless her passport expires earlier than that. If that is the case, once she has a new passport, she could apply to extend the OWP.

Regarding interviews: How many, spacing (ie. around month 4-ish...and say, month 11 or 12?), number of days/weeks notice (thinking flight bookings here), both applicant and sponsor required at all?

Flight bookings? I thought you were applying inland.
 
We are indeed, but I sometimes work in the hinterland and we will be in Thunder Bay, Ontario. (Unless I am mistaken, interviews would be in larger centers like Toronto (Mississauga), Calgary, Vancouver.)

If an interview is required, it generally means IRCC thinks there is something wrong with the relationship, i.e. Marriage of Convenience. This results in the app being transferred to the local office closest to you. If there are no local offices near you, then the interview would be scheduled at one of the temporary offices close to you. Have a look on the website.

Honestly, unless you guys have red flags, an interview is not likely.
