Family Class Sponsorship vs Working Permit

My wife is a citizen in Canada and we have been married for 12 years.
Meantime, I am also having my work permit in Canada this summer.
What should be the best and fastest way to get my PR / Citizenship?
Can I apply my PR through the Family Class Sponsorship AFTER I arrive Canada (given that I am also having my working visa in Canada as well)
