I am planing for PR with my family. I have already applied for ECA for myself. Does my wife also need to apply for ECA. Is it mandatory?

It’s not mandatory but if she does and receives and ECA it will addd additional points to your principal applicantion (PA). same goes in the case for IELTS if she gives and has a decent you will be entitled for extra points as PA

Two or more certificates, diplomas, or degrees. (One must be for a program of three or more years):

- With accompanying spouse - 119
- Without accompanying spouse - 128
