Hi,
I have done M.S from U.S and i have over 3 years of work experience in United states as a software tester [Automation]. I have come back to India and have worked in india for 2 years now.

Now I am in the process of my canadian PR. I have started my research on canadian job market. after going throgh all the posts available, i am curious to know that will my U.S. education and work exp be useful?

also what are my chances of getting the interview calls from india itself?
 
Your US experience will be helpful as Canadian employers usually consider it equal to Canadian experience. However, I don't think many will be interested in pursuing you until after you land in Canada. Given the PR process is somewhat uncertain, it's fair to say they will hire people they know can be in Canada by the start date.

Once you have your COPR and flight scheduled to land in Canada, I would begin the application process. In your cover letter, let the employer know you're moving to your prefered location in Canada X date and will be available to interview Y date (a few days later, whatever you're comfortable with). I would write a sample, but my cover letters are horrible....... :)
