Kindly anyone give me suggestions or idea that my brother is a Canadian passport holder and Permanent Resident can he apply for Sponsor to me for my PR or work permit.?

How old is your brother and his martial status? Are your parents or grandparents alive? If your mom is still living, then he cannot sponsor you for PR.

My brother is 40 years old. Yes he is married his spouse is also Canadian national and passport holder they both live there. Yes our parents are alive but my mom and dad live with me in my country.

So he cannot sponsor you as he is not a ”lonely Canadian”. You get CRS points for having a brother in Canada.

How could I get CRS points..? And one more thing, if I have a job in Canada but that company can't give me work permit because that company says that you should get work permit by yourself but we will provide you a job. So is it possible that I get work permit from any government department.?

To immigrate, you need to be eligible through Express Entry. If you have a profile, you get additional points for having a sibling in Canada.

A job offer is not enough. The company must apply for a LMIA paying $1k and proving no Canadian or PR is qualified to do the job, only you have the experience. Once you have a LMIA , you apply for a closed work permit proving you can do the job through employment letters, education assessments, language tests etc. Then with the work permit, you can work only for that company.

I will check legibility on Express Entry. I heard that people go with visitor visa and convert to work permit, but I am wondering how is it possible and how could people do that..?

You cannot covert a visitor visa to a work visa. Few people do it. You need to find an employer who is willing to pay for and go through the LMIA process. If the employer gets the LMIA then you can apply for a closed work permit. You need to show that you are experienced to do the job so it can be refused. The process can take a very long time.

Yes I am 42.
Not Master and Phd I am graduated.
I will search some employer may be they give me LMIA. I think there is only one way to search employer.

Get an ECA completed for your degree and take IELTS. Most jobs postings will say only apply if a citizen, PR or have work authorization (those on study permit, PGWP, or SOWP, IEC).

I have started IELTS and will get good band. What do you mean ECA.? I have applied for Education but I they are not giving me admission due to education gape and age.

ECA is an education assessment of your degree to show equivalency to Canadian education.
 
In general it is very difficult to immigrate after age 40. You can’t just apply to a study permit and come to Canada you must show that it makes sense for you to stop working and go back to school and that it will lead to a career advancement in your home country.
