I don't think it's a serious blocking step. Once you paid it, they confirm it right away or withing 1 day.
You should send them the receipt by email. That's the fastest way.

This reference letter is in support of Mr. XX application for Permanent Resident status of Canada through PhD stream in the Federal Skilled Worker. Mr. XX is currently registered as a full time PhD student with the Department of Chemical & Biochemical Engineering at the University of XX. d. Assisting me (Prof.XX) to further modify and verify the research results from experiments
e. Assisting me (Prof.XX) in the preparation of scholarly articles in peer-reviewed scientific journals, and preparing for research presentations and conference posters.
f. Training undergraduate summer research students on research methodologies
g. Assisting me (Prof.XX) with writing grant reports and proposals regarding to the extrusion pretreatment of corncobs for bioethanol production and discuss with me for further modification.
h. Assisting me (Prof.XX) to form guidelines for design standards and future research.

Starting from September 1st, 2011 till December 31, 2012, The University of XX had a coopering program with the XX College which was supported by an industrial company. This project was the same as Mr.XX research project. Mr. XX moved his work to XX College as a research assistant mainly using XX College’s Twin Screw Extruder equipment. Mr. XX working load there was 20 hours a week for experiments (Please see the reference letter from the XX College). My. XX stipend was paid from both University of XX and XX College during that time.

Mr. XX is contributing at least 10 hours/week towards the research assistantship from Sept 1st, 2010 to August 31, 2011 at the University of XX, 20 hours/week at XX College from Sept 1st, 2011 to December 31, 2012 and 20 hours/week at the University of XX from January 1st, 2013 to April 30, 2014. The remaining time is reserved for his own research work. The total work experience as a Research Assistant accumulated by Mr. XX at the University of XX is therefore not less than 1260 hours and 1392 hours at XX College.

In addition to full tuition remission, his annual income towards the research assistant was $4000 for 2010, $17576 for 2011, $18864 for 2012, $11712 for 2013 and $4856 for 2014.
