My brother wants to apply for Canada Visitor from US but he is on visitor visa on US. Can he apply for visitor visa from US and does he need to provide biometric?

Yes - he can apply from the US. Yes - if he's from a country that requires biometrics then he will need to go through that process. Note that it make take a few months for his application to be processed. Generally speaking, we recommend people apply while they are still in their home country rather than from the US due to the long processing times.
