I am in the process of applying for a temporary work permit visa for 3 years for myself and a tourist visa for my mother. As I want to visit USA, should I apply for USA visa from here or can I apply from Canada?

Better chance of getting a US visa in your home country than as temporary worker in Canada.
 
If he establishes significant ties to Canada and can satisfy the officer that he will leave the US, he can get a visa. Done it before with no problems. A few scenarios I know have led to rejection:

1. applying less than 6 -12 months after arriving Canada
2. student applying a few months before graduation or student not currently enrolled in school
3. worker applying shortly before contract is up or still in probationary period of employment, etc
