@nyssa I'm sure you can send it to the courier address (a few people have chimed in on this thread saying they have successfully sent their applications to the Robert Speck address), but I certainly wouldn't worry about the size of your package not fitting into their physical P.O. box. It is my understanding that the post office simply holds onto large packages for P.O. box subscribers and places a slip into the box letting the renter know that they are holding a package for them.
Plus, it would make no sense if the case processing centre's default mailing address (the P.O. box) is unable to accommodate large packages, especially considering the size of an average application. They must get boxes and boxes sent there daily.
FYI -- I "International Express" mailed our application to the Mississauga P.O. Box from NY on Friday (it fit in a large envelope for $35), and USPS tracking says it has crossed the border, made it through customs, and is currently "At Foreign Delivery Unit, CANADA" as of 8:03am this morning. Woot! ;D
