I am going to submit my application package soon. However I am slightly confused with the mailing address. It gives a regular mail address (the P.O. Box one), and a courier address:
Citizenship & Immigration Canada
Canadian Experience Class
Centralized Intake Office
49 Dorchester Street
Sydney, NS
B1P 5Z2
Canada

Question 1. What if I want to send it by Canada Post registered mail or XpressPost? Which address should I jog down?
Question 2. What would be the best option? Xpress Post? Registered Mail? UPS Store? Fedex?

If XpressPost then the PO BOX address

Xpress Post is reliable enough. If your using UPS or Fedex, remember to put in the Courier address provided instead of the P.O. box address

Inline in Red.

Use XpressPost. It is a courier service so:

Courier address
Centralized Intake Office
49 Dorchester Street
Sydney, NS B1P 5Z2
Canada

I think CPC explicitly says to submit to P.O. Box if using Canada Post: http://i.imgur.com/w0sBtZ4.png

But, it is debatable if XpressPost is a courier service or something else. Don't want to get into that :)

Xpresspost is essentially a courier service, albeit one operated by Canada Post. If you choose Xpresspost it doesn't really matter which address you choose :) I sent mine to the street address, others have used the PO Box, and in either case it gets there and is signed for.
