Wanted to know if anyone has an idea on what all cities in India are eligible for passport pick up after one gets letter to submit the original passport for Canada visa.

As of 9th July 2022, you can submit your passport and pick it up from the VFS global center in a city near you. If you still want to go for the two-way courier procedure, fill out a request via webform.
Follow the instructions, https://vfs-cic.mioot.com/forms/PS/ and at the end you'll see the space to enter your pincode (residence). That'll tell you if VFS global/BlueDart can pick up your passport and deliver it.
