Hey, I am a Post graduate work permit holder I live in Quebec and working as an assistant manager for a small local business near me.

Unfortunately, in my first year of PGWP, I was not able to find a NOC job quickly but thankfully I found one which I am working at now.

My PGWP is expiring in 1 year and 9 months, and I am trying to start gathering my documents and thoughts I am going to do a french test soon etc...

My question is can I apply for PEQ and Express my interest in staying at Quebec at the same time or is that not allowed?

You are free to give me any advice or anything that can help me.

If you want to be eligible for EE then live in Ontario as a tax resident and work in Quebec.

My question was different but your answer interest me, so I can apply for EE as long as I live in Ontario but work in Quebec.

You need to show intent to leave Québec to apply for EE. You need to live and work in Québec to apply for PEQ. The two are not compatible, you need to make a choice.

I mean the Expression of Interest program in Quebec not EE

you can apply both but why you do Express of Interest (EOI) when you're qualified for PEQ. The EOI is based on scores and you have wait for invitation to applied so it will take more than 6 months to process than PEQ.
