FSW and work permits for professionals

I have 435 points on the express entry profile (age 29 = 110 points, doctor of medicine degree = 150 points, language test = 125 points, skill transferability (foreign work experience >3 years) = 50 points).

I have also applied for Ontario PNP, still waiting to get a reply.

I am aware I need at least 450 to qualify for ITA.

How do I apply for work opportunities to boost my points? Is it possible to apply for a work permit without a job? Will the LMIA be applicable to me? (Please note that I cant yet apply for physician jobs as I still need to complete a residency program, but I would like to apply for other health related jobs)

Also, What is the process for self-employment if I want to set up my own business in medical education?

I assume you are currently outside of Canada?

If so, no - you cannot apply for a work permit without a job.

You need an LMIA and full time job offer to apply for a work permit.

There really aren't any work permits available for self employment. To obtain a work permit you would still need to secure a full time job off and approved LMIA through an employer in Canada.
