My spouse is coming on a TRV and his sponsorship PR is in process. Can he apply for a work permit once he gets here on the TRV?

No, he would require an approved Labour Market Opinion and then would have to apply outside Canada (of if eligible at a POE) for a work permit.
 
And only an employer can obtain an approved Labour Market Opinion - so the first thing he would have to do is find an employer in Canada who is willing to offer him a full time job. That employer would then obtain the LMO.

Yes he can.

If you already got the AOR then he can submit an application for an open work permit. Use the online tool to see which documents you will be needing.

Answer is still no. Even with AOR.

OP's spouse has been sponsored via Family Class (spousal sponsorship) using the outland application method. So no open work permit is available. OWPs are only granted with inland application method for spousal sponsorship.
