i am a Canadian citizen and living in Saudi Arabia. My brother is in usa right now in business status, can my brother take asylum?

No he will not be able to return to Pakistan not Saudi Arabia. He will also have to show that tried to resettle in another region of Pakistan to escape the threats. The asylum approval rate is quite low for Pakistani applicants so if he is able to immigrate or stay in the US/Canada using an economic stream I would recommend that.
