My personal view is that it should not matter since your nomination would be coming from the province and not the city, I choose Regina though.

It probably takes few days for reflection. For my case, it took around 4 or 5 working days. Hope they will reflect soon. Not to worry.

If I can recall correctly, my NOC code also got changed after getting the ITA. I would suggest you to align your NOC code with the one in nomination i.e. 2171.
