Can a person apply if they are already in Canada on work permit but not permanent residence? Should the job offer from LMIA?
Any one please can tell us about their experience?

I applied for Express Entry while on a post-graduation work permit. I didn't have a job offer, but my Canadian work experience and education credentials were enough to meet the eligibility criteria. It took me about 6 months to receive an ITA.

OP is not asking about Express Entry.

The job offer can be without LMIA, as your case that you are in Canada then i don't know the situation if you can submit.

If they're already in Canada on a valid work permit, they may still be able to make an asylum claim if they meet the legal requirements. A Labour Market Impact Assessment (LMIA) job offer isn't required just to apply for asylum, since refugee protection and work permit programs are separate. It's always best to review the individual circumstances carefully and, if possible, get advice from a qualified immigration professional. I hope someone who has gone through the process can share their firsthand experience.
