Previous employer won't include my duties

Not necessary - you can get it from any previous colleague even on plain paper and get it signed and notarized along with the LOE
A previous employer despite repeated requests to include duties won't oblige. Will CIC reject my application because of duties missing from a previous job?

Agree - no one can sign on the letterhead. Which is why they will give you a letter on plain paper in a personal capacity since they have worked with you or have been your supervisor. In any case as long as what you indicate on the letter is true, it will not be an issue. You will support this plain paper letter with proof of employment - joining letter, paystubs, etc. so it is perfectly fine.

I too am going to do the same - have obtained letters signed by current and ex-colleagues. Think of it this way - you may not want to disclose your immigration plans to your current employer. How will you get the letter in that scenario if you are using that work experience to claim points.
Actually, my ex-supervisor is not in that company now, and can he sign the letter?
Secondly, how will I get that if he is in another country? I mean, if I need to create a document or my ex-supervisor has to?
Lastly, how would he stamp if he is not in my former company?
