As far as I know (have experienced) we do get 50 points for a valid job offer. I dont think a letter from the manager would be sufficient. It should on the company's letter head with your proposed pay structure as well.

Hi, We do get an employment letter from our company and the letter clearly states that the offer starts from the day we become permanent residents of Canada. So I am not sure if you can get a new employment letter without telling them the purpose.
