What do you mean by apply to Ontario? I don't believe you have to explicitly apply to Ontario, you just need to mark as one of the provinces in your profile and it is picked automatically if your CRS > 400. Is it not the case?

And job bank registration can be started immediately after creating the profile or even for that you've to wait 24 hrs? If I re-create a new profile now, how long will it take to get it active (incl. job bank thing)? I created my profile on May 1st. On one hand I would like to recreate the profile to get NOI from Ontario but I also don't want to miss next cutoff if my profile is not active by then.

I also updated my spouse's IELTS today. How long does it take for new CRS to be updated? It still show the old one. I signed and transmitted my application.
