is the letter of reference the only thing you need to proof your work experience in canada ? is that enough as a proof ?
how the processing office check that ? by contacting the employer ?
or I need other documents to support the letter of reference ?
 
The reference letter is the only mandatory document required but nobody here can say whether it will be enough proof.

If you have your T4 or paystubs provide them.

I don't think its enough to make your case strong, you need to back your reference letter with T4 or Paystubs. You can even upload NOA for the years you have filed taxes. It makes VO works easy as well as increase your chances to get PPR sooner because if they would ask for additional doc that will delay your file. They will not reject you for Paystubs or T4's but if they would not be able to contact your employer by any chance your file lag behind.
I uploaded everything with my file and tgey didn't even call my employer for verification and i got approved.
