Has anyone submitted his full time job offer Without HRSDC's confirmation to the CIC office where his PR application is filed?

If yes, what was the CIC reaction?

Alternatively,

if I send my job offer Without HRSDC's confirmation to CIC office will that have a negative impact on my application? Will they send it back to me? Or what?

Unless you are in Canada on a TWP and the employer's letter states that once you are a PR they will employ you permanently, otherwise without HRSDC approval, it is pretty well worthless.
