You cannot apply as a PR until after you have completed your landing. Until then, you are not a PR.

Being a PR does not necessarily make the slightest difference on whether you get a US visa as your home country passport will still probably influence any result.

You can certainly apply for the Visa. Whether you will get it or not depends upon your personal circumstance. You have to show strong ties to Canada such as having an established residence, job, family etc. Then it is up to the consular officer's discretion. The longer you have been PR, the better your chances.
