Two full time work

I want to know about full time work on post graduation work permit. Can I work 80 hours a week with two different employers.i am working as a supervisor with one employee and with other employee I am working as general labour. Will it affect my PR or any work related to that?

You can work as much as you want.

Yes, a PGWP allows you to work as much or as little as you like for any employer in any job. It's an open permit, there are no conditions (unless you have something specified on your permit, which is very rare)

No, it will not affect your PR. You can work 10000 hours a week, if you want to.
