If for some reason the spouse and dependants are unable to enter Canada(and thereby become PR) within the stipulated time mentioned on the counterfoil; can a second spousal sponsorship application be submitted immediately afterwards, or should the sponsor and applicant wait for some time period before re-applying?

Contact IRCC or the embassy and ask what the best course if action would be. Maybe they can extend the counterfoil if you have serious reasons preventing you from travelling.

Yes, a new one can be submitted.

But you really should consider contacting IRCC and seeing if arrangements to extend or issue new COPR within the current PR approval is possible. IRCC traditionally did not do this often but more flexibility now with covid as government ws recommending all not to travel (and seems to be the case again now). Note, an extension might only be for three months or a year - so if it's a change of plans and you don't intend to move for a few years, might be no sense.
