Who should sign the reference letter for the work experience, HR, manager, or the president of the company?

As long as it is an authorized personnel of the company, it doesn't/shouldn't matter. The whole objective is to provide a reliable and accurate proof of your work experience claimed on the application, on the basis of which you seek immigration benefits.

Ideally your Manager or HR. In case CIC authenticates, someone should be able to confirm your work expereince
