if some one file refugee in Canada and after work permit can asylum seeker (before hearing of refugee) can apply for PR as skilled worker in Canada? If yes, then after negative hearing can asylum seeker stay in canada on basis of applied PR as skilled worker category?

If you meet the requirements, you can apply. However, it will not entitle you to remain in Canada. You will be under an enforceable removal order when your refugee app is refused, so you will need to leave Canada and will need an ARC to return.

No because during whole PR process you have to maintain legal status in Canada. if your refugee claim is rejected, you do not have legal status in Canada, so you would have to apply for TRV for example which however can be denied.
