i believe the OP meant at the time of application (while filling up the forms)

It really depends. If the applicant has dealt with CIC before, or has gotten a visitor visa or visitor record , the applicant has already has UCI.

Now if the applicant is filing for PR sponsorship for the first time in the sense of dealing with CIC, then yes the answer is no. The CIC will assign an UCI then.
