I landed last October and going to permanently move to Calgary this July. I am now in US with H1B VISA. I need to go Windsor or Hamilton in June to take the Canadian medical exam to entitle me to practice in Canada. I have PR Card already, but not physically with me, still in my friend in Canada. Not allowed to mail it oversea per CIC policy.

My question is: is there any problem for me to enter into Windsor or Hamilton in Canada with my COR copy stapled in my passport and I drive with my car? I

There is no such CIC policy. Your friend can go ahead and mail your PR card to you. Many people have done this without any issues.
 
Agree with scylla. No such restrictions exist. Ask your friend/relative to fedex it. You should get it in a day or two. Avoid normal post as these are important docs.
