What is the difference between job offer and LMIA? I am in a Post graduation work permit for 3 years. I have a full time permanent job in Bookkeeper position in Toronto. Do I need LMIA from the Employer?

Yes.

If you want to claim the 50 job offer points under Express Entry, your employer must obtain an approved LMIA.

This is because you are on an open work permit with no employer named.
 
Thank you. I finished my study in Ontario. If i apply through Express Entry and select the provinces I am interested in, Do I still need to apply through PNP system of that specific province?

If you have enough points to be selected through Express Entry directly, then you're all set. You would want to have 470+ points to be in good shape for this. How many points do you have?

Your employer can select #1 to avoid the fee. They will still need to advertise the job for at least one month to prove no Canadian could be found for the role before they can apply for the LMIA. Make sure they read the advertising requirements in detail. They will need to advertise in three different locations and make sure they pick options that meet the LMIA advertising criteria. You also want to make sure you're being paid at least the median wage for your NOC and location.

They advertised through Indeed. What other province can I expect with 428 points? I have ECA for bachelor degree 3 yrs. and also Post graduate diploma for 2 years in Canada. In that case , Shall I select that I have two or more degree when CRS calculation?

They need to advertise again if you want to move ahead with the LMIA. Advertising through Indeed alone is not sufficient. Also, the advertisement they placed to hire you is too old to be used in the LMIA application.

Yes, you can select two or more degrees for CRS.

In terms of other provinces, you'll need to research that. I would focus on increasing your score since it will likely be difficult to be selected without a job offer in that province.
 
I don't know how I can increase my score, I tried several times IELTS GT and getting 6.5 most of the time. Can u advise what other way I can increase my score?
 
I have 8 years job experience back home. But I think they don't count experience more than 3 years

Improving your language score is the easiest way. Everything else is generally much harder. I would recommend that you focus on your language score.
