If you have enough points for FSW, you just need an AEO (job offer) and then you can apply. The job must be skilled though.
If you do not have enough points for FSW but you have an employer who wants to hire you, it is possible that you can apply through PNP directly depending on the province or the employer can try for an LMO which would allow you to apply for a work permit. If you get it, you could have options to apply for PR later on. Preferably the job also has to be skilled for that too but there are a couple of provinces that offer options for certain low skilled jobs too.
