I am applying a visit visa for my brother online and there is a question that bothering me is that "Do you have a family member who is a Canadian citizen or permanent resident and is 18 years or older? "

When I checked the Glossary on the website it says "

An applicant's closest relatives, in the context of an application to IRCC. It is defined as a spouse or common-law partner, dependent children, and their dependent children.
"
I am a Canadian Citizen, can he say Yes to this question (for me as a brother)?

No, as per the glossary you are not a family member which means his immediate family unless you are a dependent of his which seems unlikely.
