I came into Canada from Nigeria to do Licensed Practical nurse examination on a visitor status which i have renewed and will expire end of March 2014. I passed my LPN examination and now registered as LPN nurse however I have a positive LMO as a live in-care giver in Calgary. Can i go to the border with my documents in Southeast of Calgary at Couttis, Alberta/Sweetgrass to get work permit instead of returning to my country? What are my chances of getting the work permit? How long will the process take?

Nope, all LCPs must apply to an overseas office.

Alright thanks.I understand one can apply through US since it is an office outside Canada, do they request for interview or medical test? If yes do they allow visitors to enter US?

An interview may be required. It's up to the US if you are granted a visa or not - and just because Canada has asked for an interview doesn't mean the US will approve your visit. You also don't get much notice for the interview. So if you don't already have a multiple entry visitor visa for the US, I wouldn't apply through there.
