I am currently on an international Canadian experience working holiday visa which expires in June. This is only valid for 2 years. We're both British citizens.
My husband (was just boyfriend when we moved here 2 years ago) is a permanent resident. We've been together 6 years.
If he was to sponsor me now can I still work in my current job while the application is being processed? It's unskilled unfortunately, I work as an accounts receivables clerk for a golf club.

No, you won't be able to work.
He can sponsor you outland even if you are in Canada, and you will have to stop working once your permit is over, unless you find an employer willing to give you a LMO (difficult, and unlikely for non-skilled professions). You can stay in Canada though, by applying to change your status from worker to visitor, and wait out the rest of the PR.
IF you file soon enough, you might not have to wait too long... london is a "fast VO", stage 1 is currently listed as 55 days but a lot of people get done faster, and for straightforward cases, London is somewhere between 3 to 5 months.

If you apply inland, you can have "implied status" which means that you are allowed to stay in Canada ( even without applying), but your work permit won't be extended ( at least that seems to be the case for people on the forum) as you have an IEC visa - not extendable.

There are very few cases where it is more favourable for visa-exempt to apply inland rather than outland, and I don't think you're one of them, so outland would probably be a faster way to go.
good luck,

Thanks for the response!
So how do we apply Outland if we are here?

It's a bit confusing, but outland refers to which application will handle your application, not where you actually are.
So if you are outside of Canada, you can only apply outland.
If you're in Canada, you can choose to apply inland ( through Vegreville), or outland (first stage is Mississauga, second stage in your case through London). It's completely legal, lots of people do it, and it's "encouraged" by CIC (it's somewhere in the Inland guideline: if you are in Canada but can benefit from faster processing by applying through Outland, please do so, or something similar).

For outland: just go to CIC website, click on "sponsor your family", and follow the link which says " sponsoring your family from outside of Canada".

As your partner is a PR ( and not Canadian), he NEEDS to be in Canada while sponsoring you. But in your case, you could be anywhere in the world (including in Canada).

Hope it helps!
Good luck,
