I am the secondary applicant & my elder brother is citizen of canada.
Express Entry online submission expects you to let them know whether any family member of secondary applicant is canadian citizen.

Is this entry provides any benefit to the application at later stage, in terms of:
Proof of Funds relaxation?
adaptability?
or any other factor?

As far as I am aware, the province of Manitoba has a stream called Manitoba Support Stream. You can find more details here http://www.canadavisa.com/eligibility-requirements-manitoba-provincial-nominee-program.html#Manitoba Support Stream. In addition to other eligibility criteria, the basic criteria is that the Manitoba Supporter should be a Canadian citizen or Permanent Resident and also he should be able to support your Settlement Plan.

I have one question, who is the primary applicant, since you have mentioned that you are the secondary applicant?

I am secondary applicant & my spouse is primary applicant. My elder brother is citizen of Canada & is well established there. My CRS is 458 and thus I am hoping to get direct ITA in coming draws. However, I was exploring the alternatives of proof of funds or any other factor which may be benefitted due to the elder brother of secondary applicant being citizen of Canada.

No - no benefits later. You have to show the same proof of funds as anyone else.
