Before applying I want to clear some points from seniors.

1-I have not taken IELTS test so I will mention that I have not taken test. But I will claim for maximum points. Result will be sent lateron after first approval. Is it will return application without process or I will be able to get initial approval and will send ielts result to visa office for further processing.

2-I have applied for passport. Passport will be achieved after 2 to 3 weeks But I have a computerised Visa application receipt from Passport office. Will it be suffice to send application on urgent basis. I will be able to send passport photocopy to visa office. Will the application will be returned without processing or it will get initial approval.

3-May I claim education points of my spouse later on to visa office by submitting my spouse fee. I may need my spouse points in case of lower band in IELTS test to get passing marks of 67.

4- I have two master degrees from Pakistan. My first master degree in Physics was obtained after 16 years of full time study. Later on I achieved my second master degree by studying two years full time. So total study time is 18 years. So how much I claim points 25 or 22.

5- I have contiuous 8 and half years experience by getting three promotions from supervisor to Deputy manager. Responsibilites increased and somewhat changed after each promotion. Will I need a reference letter of three postions or I can show only four year experience with specifice management duties.

You can take IELTS later but it is recommended that you do it before applying so that you know your total points. Applicants leave it until the end and they dont get the desire marks and in a result have to withdraw their applications. If you send it now thats fine they will processs your application.

You can write "applied for" in the forms and send it later.

If you add her in your application now you will receive points for her education.

I beleive you will get only 22

As you have three position in the same company you can get all of them written on the one letter and explain your job duties for the position you are applying for. You need minimum of 1 yr experience to apply.
