I am sponsoring my husband who has a child from his PREVIOUS RELATIONSHIP. We declared his son in our application and now, the embassy is requesting us to let his child from his previous relationship to have his medical. However, we have no idea about the whereabouts of his child nor his ex wife. We never had seen them nor heard any news from them since on...

Please enlighten us on what should we do. Any help is greatly appreciated :)

You could search for the mother and child (facebook, private detective, contact former friends/acquaintances etc.etc.) and see if the mother will allow the medical to be taken. Generally people are not that hard to find.
Otherwise you need to inform the consulate of the situation and prepare and sign a notarized separation statement which says that you understand the child is not a member of the family class and can never be sponsored.
I would start searching and use the separation statement only if all else fails. The VO will want to see that you did make attempts to locate the child and get the medical done.
