Question about time to apply

Hi,

1. I would have enough one year experience on this June and be eligible. Therefore, should I create profile before then one or two months because it will take time to be invited?

2. Can reference letter from my manager prove that my work qualifies for NOC?

1 - No. You shouldn't create your profile until you actually meet the minimum entry criteria.
2 - Yes, you will need a reference letter following IRCC's guidelines to prove your work experience.
