If by job letter you mean a LMIA, you've got a pretty good chance of receiving an ITA in the next draw after you create your Express Entry profile and enter the pool.

We cannot predict when the next draw is going to occur, but they usually happen on fridays.

As you already have a LMIA, if you qualify for one of the programs under Express Entry and prove being able to provide all the evidences CIC requires, it is very likely that you will get your Permanent Residency Visa.
