Hi I am @CRS: 451 and received expression of Interest from Ontario. Have some questions about it:-

1) Is it guaranteed that after receiving EoI, I would get The provincial nomination with my profile or it is still a matter of luck? I have all the documentation they are asking. Do they grade the profiles or if you get EoI, and all he documents in place, you would get it?

2) What are the timelines for receiving the PNP nomination? Does it increase the total timeline or the total timeline stays the same?

1. If you meet all the requirements and you submit a complete application, you will be nominated. No grading.

2. 6-8 weeks. On average, you should expect it increases the timeline. Considering you aren't sure when the draw would get to 451, you may want to proceed with OINP.

My current employer is not willing to provide reference letter for job duties which covers around last 4 years of my experience. I have reference letter on company letter head from my previous employer for another previous 5 years.

Could you please help me if possible, if OINP would accept Job duties letter on affidavit from a senior colleague for my current employer. I could provide an email from my company HR that they could not issue these letters from their side.

I do think it would work. In addition to the email from HR, perhaps you could also try to provide any secondary documentation possible: contract/promotion letters, pay slips, etc. If your senior colleague can also provide evidence of his employment (company ID card, business card, etc)...it would help.
I've seen testimonies of candidates whose offices declined providing the ref letters. It should be fine.
