If the PA on an application hasn't got an AEO or LMO, would it make a difference if the spouse had one instead?
(sorry if this is already been answered but after a half hour search I couldn't find a similar request!)

Nope.  It is the principal applicant the CIC is interested in.  If the spouse had worked in Canada for 1 year, you would get 5 adaptability points.
