Hi, everyone.

Must I first enter Canada at or near the location I indicated as my intended destination on my application form? I indicated Toronto on my application form, but I am planning to land in Vancouver.

By the way, can anybody tell me what the job market is like in Vancouver and Toronto?

You can "land" where ever you want as long as it not Quebec.  If you were accepted as a PNP in a province you should check the info that came with the PNP in establishing yourself in the province that accepted you.
