How can I apply for work permit while here in visitor status

Can someone tell me if I can apply for my temporary work visa from inside of Canada while I am here on Visitor status?

I have a positive LMO and all my documents ready to go.

You can send your initial application to the US, either Buffalo or Seattle.

I am currently under implied status in Canada and I have applied for a WP out of buffalo. I have received the letter of authorization as well as the W-1 visa(TRV) in my passport. Do I still have to leave the country to activate the work permit/get the work permit issued or can I just go to the Niagara Falls Border and get have the work permit issued there? Your help is greatly appreciated.
