There is no 2 year condition that I know of. For your PR, it is not very clear cut. You only sign a statement stating that you intend to settle in Manitoba. When you promise that you intend to do something, it is not a lifetime committment. If you land in Manitoba and stay for several months and try to find work but you end up not finding anything and move to another province, nobody will hold that against you and nothing will happen to your PR.
For your MPNP sponsor however, he signs a statement stating that he will help you settle in MB and if you do not settle successfully in MB, he can not sponsor anybody else. That is a lifetime thing for him. Say you settle in MB and your relative then applies to sponsor another relative. MPNP will ask him, who have you sponsored before and where do they live. If you are still living in MB, then he can sponsor another person. If you have moved, he can not.
